I don't think LNP is awesome but it is a good and safe delivery platform. I don't like LNP for RNAi delivery because it is IV and it has to be given with steroid premedication. Also, it requires frequent infusion which is not practical and cost prohibitive. But it also has its pluses such as it can be used for the delivery of mRNA and for Gene editing where GalNAc doesn't work. It is ideal for Gene editing delivery where only a single or limited number of injections are required. NTLA and SGMO are using LNP delivery.
Advantage of GalNAc is it is SubQ and doesn't require steroid premedication. It is also long lasting - quarterly and bi-annual SubQ injections are easily attainable. I expect all of the future ALNY drugs based on their ESC+ delivery will have a minimum of quarterly injection. I won't be surprised if some of them require only annual injection based upon the drug and KD required.
GalNAc has been proven and I believe AB-729 will be successful. We will see in a year from now.
